Successful peptide mapping requires selection of the right cleavage enzymes. Protease V8 (also known as endoproteinase Glu-C) complements tryptic digestion, specifically cleaving peptide bonds on the C-terminal side of glutamic and aspartic acids. Protease V8 from MP Bio has been validated by more than 200 scientific publications and provides:

  • Robust activity with greater than 915 units/mg protein
  • Specific cleavage at glutamic acid residue in ammonium bicarbonate at pH 7.8, ammonium acetate at pH 4 and specific digestion at both glutamic and aspartic acid residues in phosphate buffer at pH 7.8

Protease (S. Aureus) V.8

Protease V.8 specifically cleaves peptide bonds on the COOH terminal side of either aspartic or glutamic acids. In the presence of ammonium, the specificity is limited to glutamic sites.
